摘 要:本实验以豆渣中提取的膳食纤维为原料,进行半纤维素多糖的提取工艺研究。在单因素实验基础上,选择碱料比、提取温度、提取时间以及碱浓度为自变量,采用二次旋转正交回归试验设计结合SAS分析方法,确定提取半纤维素粗多糖最佳条件为:V(10%NaOH):m(膳食纤维)=21.2:1,温度为35.5℃,时间为5.3h。验证结果提取率为30.21%。由此工艺得到的半纤维素粗多糖采用EDAE-Sepharose Fast Flow离子交换柱色谱法进行纯化,分离得到组分HCLB-I和HCLB-Ⅱ,相对分子质量分别为4.723×107和2.996×107。The separation and purification of the hemicellulose polysaccharide from soybean dietary fiber was studied in this paper. The effect of the ratio of NaOH to material, extraction temperature, extraction time and the concentration of NaOH on the yield were studied by twice rotation orthogonal regression test design and SAS software. Optimal conditions for extraction of hemicellulose polysaecharide were shown as follows: extraction temperature of 35.5℃, extracting time of 5.3 h, and the ratio of-10% NaOH to material of 21.2. Under these conditions, the yield of the polysaccharide is up to 30.21%. Crude hemicellulose polysaccharide was then purified by EDAE -Sepharose Fast Flow, and be separated to two fractions , HCLB-I and HCLB-Ⅱ , with the molecular weights being of 47.23 million and 29.96 million respectively.
