摘 要:通过浓硫酸显色、TLC分析、紫外扫描及高效液相色谱等技术对传统发酵豆渣食品中分离出的目标菌株粗壮脉纹孢菌(Neurospora.crassa)所产类胡萝卜素进行定性及定量检测,结论如下:豆渣经粗壮脉纹孢菌发酵后,类胡萝卜素含量大大增加,且主要为β-胡萝卜素,紫外扫描测得孢子中β-胡萝卜素含量达158.50μg/g孢子,产量为8.06μg/g培养基干重;高效液相色谱检测得含量达140.98μg/g孢子,产量为7.14μg/g培养基干重,两者结论基本一致。The methods of Chromogenic reagent,TLC, ultraviolet spectrometers and HPLC were used to research the carotenoids by Neurospora crassa in this experiment. The objective strain was screened from a traditional soybean residue in Jiangxi Province.The results were as follows:there was no carotenoid in soybean residue.The amounts of the carotenoids in the soybean residue would be increased greatly after fermented by Neurospora. Crassa.And most of the carotenoids were β-carotene.The UV-visible spectrum and HPLC analysis showed the content of β-carotene was 158.50μg/g and 140.98μg/g spores and the yield was 8.06μg/g and 7.14μg/g culture medium, respectively.The results of the two methods were similar.
