摘  要:为开发微生物油脂资源,从土壤中筛选出1株油脂含量为13.52%的曲霉,经紫外线诱变选育出1株油脂含量为18.75%的高产油脂曲霉.通过对菌体产油脂因素进行单因素和正交试验,得出摇瓶发酵培养最适产油脂条件为:以30g/L马铃薯淀粉为碳源,以0.897g/L硫酸铵为氮源,碳氮比为80∶1,培养温度28℃,pH6.5,摇瓶发酵120h.验证性试验结果表明,在该条件下所得菌体生物量为12.62g/L,油脂产量为4.748g/L,菌株油脂含量为37.62%,淀粉利用率为78.53%.Screening a Aspergilli with lipid production of 13.52% from soil, a mutant of superior Aspergilli was obtained with high lipid production of 18.75% through ultraviolet mutagenesis in order to empolder microbial oils resource. The optimum shaking flask fermentation conditions for producing lipid were optimizedey investigating single factor and orthogonal test influencing on the lipid accumulation: carbon source is 30 g/L potato amylum, nitrogen source is 0.897 g/L (NH4)2SO4, C/N is 80 : 1, temperature is 28℃ ; pH 6.5. Shaking flask fermentation time is 120 h. As a result, strains biomass is 12.62 g/L, lipid output is 4.748 g/L, lipid content is 37.62%: starch using ratio is 78.53%.
