摘  要:以羟自由基(OH·)、超氧阴离子自由基(O-2·)的清除率和酪氨酸酶抑制率为研究对象,研究从富含胶原的海产品下脚料中提取的海洋活性胶原肽(Marine active collagen peptides/MACP)的体外活性,并进行初步分离。结果表明:MACP的OH·清除率(IC501.42mg/mL)高于谷胱甘肽(IC508.92mg/mL)和VC,低于抗氧化剂TBHQ(IC500.375mg/mL)和茶多酚(IC500.382mg/mL);MACP的O-2·清除活性(IC5014.40mg/mL)低于茶多酚(IC504.05mg/mL)和谷胱甘肽(IC500.49mg/mL),却高于TBHQ(IC5017.52mg/mL)。MACP抑制酪氨酸酶的IC50为17.06mg/mL,为常用美白剂——熊果苷的1/70。酶解液经截流相对分子质量分别为10000、4000、1000的超滤膜分离,其活性随相对分子质量的减小而增大,且清除自由基和抑制酪氨酸酶的最大活性相对分子质量都集中在1000以下。对相对分子质量小于1000的MACP成分进行葡聚糖凝胶DEAE-A25阴离子交换分离,得到2个洗脱峰,且活性成分主要集中在第1个峰上。将MACP应用于功能食品或美白化妆品中开发前景良好。The vitro activities including the ability of scavenging OH. and O2^-· and the inhibition rate on tyrosinase had been investegated and the active ingredient had been separated in the paper. The results showed that the OH. scavenging ability of MACP(IC501.42 mg/mL) was higher than GSH(IC50 8.92 mg/mL)and VC, however, it was lower than TBHQ(IC50 0.375 mg/mL)and tea polyphenols(IC50 0.382 mg/mL). At the sametime, the O2^-· scavenging ability of MACP(IC50 14.40mg/mL) was much lower than GSH(IC50 0.49 mg/mL), but which was higher than TBHQ(IC50 17.52 mg/mL). Moreover, the IC50 of MACP of the inhibition on tyrosinase was 17.06 mg/mL, which was 1/70 of the general whitening reagent abutin. It was found that the activities contain the ability of radical scavenging and tyrosinase inhibition of MACP were increasing with the molecular weight decreasing after through the ultrafiltration membrane, the molucular weight of higher active MACP was distributed under 1 000. Two active peaks were gotten, after under 1 000 fraction was isolated by iron exchange column of Sephdex DEAE-A25, both the ability of radical scavenging and tyrosinase inhibition almostly distributed on the first peak. There are good perspect applying MACP on functional foods or whitening cosmetic.
