摘  要:通过7种酶对柞蚕丝素蛋白水解的实验表明,酸性蛋白酶水解蚕丝素的水解效果最好,并经单因素及正交实验确定酸性蛋白酶水解丝素的最佳条件为:加酶量为8000u/g,pH3·8,料液比为1∶200,50℃下,水解4·5h,所得水解液中氨基肽氮含量为807μg/mL,最终得到的水解度为16·8%。经SephadexG-25葡聚糖凝胶分离出两种组分,其分子量分别为2959、1432Da,并测得其清除·OH自由基能力分别为67·52%、14·76%。Seven kinds of enzyme were used to deal with silk fibroin protein respectively. Acid protease was the best. The optimum conditions of acid protease hydrolysis were determined that the amount of enzyme 8000u/g,pH3.8,at temperature of 50℃,liquid ratio of 1∶200 for 4.5h. The content of amino nitrogen was 807μg/mL in hydrolyzation,finally the degree of hydrolysis was 16.8%. Then it was isolated two groups by Sephadex G-25,and the molecular weight was 2959,1432Da which the cleaning of hydroxy radical were 67.52% and 14.76% respectively.
