摘  要:为确定具有抗癌活性大豆多肽的分子量,对大豆抗癌肽粗提物进行超滤,采用MTT法(四噻唑蓝比色法)对得到的不同组分超滤液进行抗癌活性检测,初步确定分子量在5000~10000u范围的组分具有最高的抑癌活性,大豆抗癌肽的IC50为0.47mg/mL。用葡聚糖SephadexG-75凝胶柱对该组分进行进一步分离,收集各洗脱组分并进行抗癌活性研究。结果显示:组分A4具有最佳的抗癌活性,其IC50为0.37mg/mL。根据分子量标准曲线,得到其平均相对分子质量为6723u。To determine the molecular weight of soybean peptides with anti-cancer activity,the extraction of soybean anti-cancer peptides was ultrafiltrated.MTT(thiazole blue four) assay was used to detect the anti-cancer activity of different components obtained by ultrafiltration.Components of soybean anti-cancer peptides with molecular weight ranging from 5000 to 10000u had the highest anti-cancer activity,and the IC50 of soy anti-cancer peptides was 0.47mg/mL.Gel Filtration Chromatography(Sephadex G-75) was applied to further separate and collect all of fractions in order to research the anti-cancer activity.The results showed that:A4 components had the best anti-cancer activity,and its IC50 was 0.37mg/mL.According to standard curve of molecular weight,average molecular weight of A4 was about 6723u.
