摘 要:采用碱性过氧化氢法提取豆渣碱溶性多糖,但提取出的豆渣粗多糖中蛋白质含量较高,因此本论文对去除豆渣碱溶性粗多糖中蛋白质的最佳方法进行了研究。以蛋白质脱除率和多糖损失率为检测指标,在单因素和正交试验的基础上,比较了酶法、TCA法、Sevage法、酶法结合TCA法、酶法结合Sevage法的脱蛋白效果,最终确定了去除豆渣粗多糖中蛋白质的最佳方法为酶法结合TCA法。结果表明,酶法结合TCA法去除豆渣多糖中蛋白质的最佳条件为:酶解温度为45℃,酶解时间为2h,酶解液pH为8.0,加酶量为1.5%(以底物计),TCA最终浓度为1.5%(w/v)。在此条件下,蛋白质脱除率为91.80%,多糖损失率为35.64%。应用此法脱蛋白,蛋白质脱除率很高,多糖损失率相对较低,是去除多糖中蛋白质的最佳选择。Bean dregs alkali soluble polysaccharides were extracted with alkaline hydrogen peroxide. However, the ex- tracted bean dregs coarse polysaccharides contained a great amount of protein. Therefore, the optimum method of remo- ving proteins from bean dregs alkali soluble crude polysaccharides was studied in this paper. On the basis of the removal rate of proteins and the loss rate of polyscccharides, several methods were compared including enzymatic method, tri- chloroacetic acid method, Sevage method, combination of enzymatic and trichloroacetic acid method. The results showed that the combination of enzymatic and trichloroacetic acid method was the best. The optimum conditions was de- termined by single factor and orthogonal test. The result showed the removal rate of proteins was 91.80% and the loss rate of polysaccharides was 35.64% when extracting conditions were as follows: enzyme solution temperature 45℃, enzyme solution time 2h, enzyme solution pH8.0, enzyme dosage 1.5% (according to the substrate) , the final con- centration of TCA 1.5% (w/v). This method is the best choice in removing proteins from polysaccharide with high protein removal rate and relatively low polysaccharide loss rate.
