摘 要:以深海明太鱼内脏为原料,应用酶解法从中提取鱼油,以鱼油提取率为指标,选择合适的蛋白酶。研究了液固比(W/F)、加酶量、温度、pH值和时间等5个因素对鱼油提取率的影响并以鱼油提取率为指标,通过正交优化试验设计,获得以中性蛋白酶酶解法提取鱼油的最佳酶解工艺条件:液固比(W/F)0.5∶1,酶添加量4 000U/g,酶解温度55℃,酶解时间2h,酶解pH值为7.0。利用此工艺鱼油提取率为69.38%,理化指标符合SC/T 3502-2000的粗鱼油二级标准。Theragra chalcogramrna visceral was treatd as experiment material. The process parameters af- fected on the extraction rate of fish oil were studied regarding the extraction rate of fish oil as the index and choosing the suitable protease. Five factors which included the rate of material to water, temperature, time, pH value and the amount of enzyme were analysed on the impact of oil extraction rate. Regarding the ex-traction rate of fish oil and the organoleptic characters as the index,the optimal process parameters of neu-trase hydrolysis were obtained by orthogonal experiment,the rate of water to material (W/F) 0.5 : 1,the amount of enzyme 4 000 μ/g,the temperature 55 ℃ ,hydrolysis time 2 h, hydrolysis pH value is 7.0,under the conditions,the extraction rate of fish oil was 69.38% ,the physical and chemical properties of crude fish oil accorded with the second grade for SC/T 3502-2000.
