摘  要:利用湿筛法从小麦面粉中提取水不溶性阿拉伯木聚糖 (WUAX)进行酶解试验。对于单纯的WUAX体系 ,在较高的木聚糖酶水平下 ( 1.0× 10 - 3酶 /底物 )作用 1h ,WUAX可显著降解 ;降低酶用量 ( 0 .1× 10 - 3 酶 /底物 ) ,WUAX的酶解程度下降 ,并在反应初期表现出粘度变化的复杂性。对于 (WEAX +WUAX)复合体系 ,WEAX的存在使WUAX的酶解增溶程度下降。在 0 .1× 10 - 3 的酶用量不同作用时间下 ,单纯体系比复合体系的增溶程度高 10 %~ 2 0 % ,而WEAX的降解则基本不受共存WUAX的影响。由(WEAX +WUAX)的凝胶过滤图谱可知 :WUAX中亦含有糖蛋白 ;随酶解程度的进行 ,WEAX显著降解 。Water-unextractable arabinoxylan (WUAX)isolated from wheat flour by wetsieving was treated with xylanases in vitro.For a simple WUAX system,WUAX was markedly degraded by the xylanases at a high ratio of enzyme to WUAX (1.0×10 -3 g/g) in 1 hour.At a low enzyme dose (1.0×10 -3 g/g) the extent of degradation decreased, and the viscosity increased in the initial stage and declined then.In a mixture of WEAX and WUAX,the enzymic solubilization of WUAX reduced by 10-20%(at 0.1×10 -3 g/g) comparing with that in the simple system.The degradation of WEAX was hardly influenced by the presence of WUAX.The results from the gel filtration Chromatography of the mixture of WEAX and WUAX and its enzyme degraded products showed that some glycoprotein was in WUAX.With further enzymic degradation,WEAX was markedly degraded,and the glycoprotein was also partially degraded.
