摘  要:对菜籽多糖的碱法提取工艺条件进行研究。原料预处理后,选取NaOH质量分数、提取温度、提取时间和液料比为变量,以多糖得率为指标,进行单因素试验,考察以上4个因素对多糖得率的影响。在此基础上,选取NaOH质量分数、提取温度、提取时间为变量进行3因素3水平的Box-Behnken中心组合试验,并通过响应面分析得到菜籽多糖碱法提取的最佳工艺条件为NaOH质量分数4.1%,提取温度86℃,提取时间3 h,液料比25∶1,在此条件下多糖得率为5.86%。In this work, after the pretreatment of rapeseed and sodium hydroxide (NaOH) solutions used as extracting agent, the extraction conditions of rapeseed polysaccharides are optimized by response surface methodology. The effects of NaOH concentration, extraction temperature, extraction time and liquid-solid ratio on polysaccharide extraction yield are determined through single-factor experiments. On this basis, response surface methodology along with Box-Behnken design is applied to optimize the extraction conditions. Three independent variables are NaOH concentration, extraction temperature and extraction time. The results show that the optimal conditions of rapeseed polysaccharides are NaOH concentration 4.1%, extraction temperature 86 ℃, extraction time 3 h and liquid-solid ratio 25 : 1. Under these conditions, the extraction yield of rapeseed polysaccharides is 5.g6%.
