摘  要:采用物理离心方法获得蜗牛分泌液,分别用考马斯亮蓝G-250法和苯酚-硫酸法测定粘液蛋白质和糖类含量,用DPPH和ABTS法测定蜗牛粘液体外抗氧化活性及其热稳定性。结果显示,蜗牛粘液中含水量为95%~99%,蛋白质含量为5.0~5.1 mg/g,糖类含量0.3~0.6 mg/g。蜗牛粘液对DPPH自由基的最大清除率为20.56%。粘液对ABTS·+自由基的清除率随浓度的增加而增加。通过对粘液抗氧化功效对温度的稳定性的测定,发现粘液的抗氧化活性在20~80℃范围随温度的变化不大。当在90℃作用10 min,粘液的抗氧化活性变化不大;作用20 min时,蛋白质结构变性析出,粘液的抗氧化活性降低;100℃时粘液清除ABTS·+清除率降至其常温下清除率的56.8%。研究结果表明,蜗牛粘液的抗氧化物质可能由两部分组成:对热温度不稳定的蛋白类物质和对高温较稳定的非蛋白类物质。本研究结果表明蜗牛粘液对抗氧化活性具有积极影响。The snail secretion was extracted by Centrifugal method. The content of protein in the mucus were deter- mined by Coomassie Brilliant Blue G-250 and carbohydrate was determined by the phenol-sulfuric acid method respec- tively. Vitro-antioxidant activity of mucus was determined by DPPH and ABTS method and the thermal stability of the antioxidant activity was also determined. The results show that the water content of the mucus was about 95%-99%, with protein 5.0-5.1 mg/g and carbohydrate 0.3~0.6 mg/g. According to the results of antioxidant activity, the biggest radical clearance rate of DPPH. was 20.56%. Clearance rate of ABTS.+ free radicals was increased with increasing concentration. Thermal stability of antioxidant activity was determined when temperature was 20 to 100 ℃. Results show that the antiox- idant activity changed little in the range of 20-80℃, while 90 ℃ for 20 min, it decreased due to protein denaturation. When temperature went to 100 ℃, clearance rate fell to 56.8% that of at room temperature. The study show that snail mucus antioxidants may be composed of two parts, the unstable protein substances in hot temperature and non-protein substances stable in high temperature while both the components constitute the antioxidant part of the mucus.
