摘  要:采用响应曲面法研究碱液质量分数、浸泡时间和浸泡温度对刺麒麟菜中提取的ι-卡拉胶的产率、黏度、凝胶强度的影响。在单因素试验基础上采用Box-Benhnken中心组合试验,以碱液质量分数、浸泡时间和浸泡温度为影响因素,以产率、黏度和凝胶强度为响应值建立二次回归方程,通过响应面分析得到优化组合。结果表明,碱处理优化工艺参数为碱液质量分数12%(120 g/L),浸泡时间6.12 h,浸泡温度32℃,在此条件下提取出的ι-卡拉胶产率为50.16%,凝胶强度为308.76 g/cm2,黏度为174.50 mPa·s。在原有工艺的基础上,增加了蒸发水分的工艺过程,可减少酒精用量。经测定蒸发水分对得到的ι-卡拉胶的黏度、凝胶强度、硫酸基含量的影响不显著。The influences of 3 factors including alkali concentration, soaking time and temperature on gel strength (GS) , viscosity and yield ofι-carrgeenan extracted from Eucheuma spinosum by response surface methodology (RSM) are studied. On the basis of single-factor investigation, Box-Benhnken central composite experiments are carried out to build the quadratic regression models for GS, viscosity and yield with the above 3 factors, and the alkali processing technology is optimized by response surface analysis. The optimizing process parameters are: alkali concentration 12%(120 g/L) , soaking time 6.12 h, soaking temperature 32℃. Under the optimal processing condition, yield ofι-carrgeenan, the GS and viscosity are 50.16%, 308.76 g/cm2 and 174.50 mPa·s, respectively. On the basis of the original technology, added the evaporation process, can reduce the amount of alcohol, and it is not significantly affected on the nature ofι-carrageenan.
