摘  要:采用高效液相色谱法对肉鸭表皮组织中的松香酸含量进行检测。肉鸭表皮组织中的松香酸用乙腈提取,经C18固相萃取柱净化,采用高效液相色谱-紫外法进行检测。色谱柱:反相C18柱(250mm×4.6mm,5μm),流动相:0.003mol/L磷酸溶液-甲醇(8:92,V/V);流速:1mL/min;检测波长:240nm。结果表明:方法线性范围为0.1-10mg/L,检出限为0.10μg/g,定量限为0.33μg儋。在0.5-50μg倌的添加范围内回收率为82.2%-88.0%。实际样品分析结果显示,松香脱毛处理的肉鸭表皮中松香酸含量高达14.52μg/g。本实验建立的分析方法简便快捷、具有较好的灵敏度和可靠性,可用于肉鸭表皮组织中松香酸的定量分析。A high-performance liquid chromatographic (HPLC) method was established to determine abietic acid in duck skin. Abietic acid in duck skin samples was extracted with acetonitrile, followed by purification with a C18 solid-phase extraction cartridge, and determined by HPLC with an ultraviolet (UV) detector. The chromatographic conditions were established using a C18 column (250 mm×4.6mm,5μm) with a mobile phase consisting of 0.003 mol/L phosphoric acid- methanol (8:92, V/V) at a flow rate of 1.0 mL/min and the detection wavelength was set at 240 nm. Results indicated that the linearity ranged from 0.1 to 10.0 mg/L, the detection limit of abiefic acid was 0.1 μg/g, and the quantification limit was 0.33μg/g. The recoveries from spiked samples at concentration levels of 0.5-50 μg/g were in the range of 82.2%-88.0%. The developed method was successfully applied to the determination of abietic acid in duck samples epilated with colophony. The content of abietic acid in the duck sample could reach 14.52 μg/g. This method proved to be of high maneuverability as well as excellent sensitivity and accuracy, and could be used for quantitative analysis of abietic acid in duck skin tissue.
