摘 要:探讨具有降血糖作用的南瓜寡聚糖的分子组成、分子结构信息与其活性间的构效关系。以南瓜粉为原料,经水提取、有机溶剂分步萃取和柱色谱分离纯化,得到1种水溶性寡聚糖(PCE-CGL)。经高碘酸氧化、Smith降解,采用IR,NMR等方法对该多糖的化学结构进行表征。结果表明,PCE-CGL分子质量为5.030×103u,主要由葡萄糖、半乳糖和肌醇组成,物质的比为6∶1∶2,并含有少量的氨基糖。主链主要是以β-1→4糖苷键及β-1→3糖苷键连接的葡萄糖、半乳糖、葡萄糖胺及肌醇,侧链主要是以β-1→2糖苷键及α-1→6糖苷键连接的葡萄糖。提示PCE-CGL很可能作为胰岛素化学介体的类似物,通过增加化学介体的数量来改变胰岛素敏感的酶的活性,从而实现其生物学效应。The aim of this paper was to research the molecular composition and the structure of pumpkin oligosaccharide with the hypoglycemic effect and the structure and activity relationships(SARs). An water-soluble oligosaccharide(PCE-CGL) was extracted by water extraction, and organic solvent extraction step by step, and further purified by column chromatography from pumpkin powder. The structural characterization of PCE-CGL was confirmed by NaIO4oxidation, Smith degradation, IR and NMR techniques. The results showed that PCE-CGL contain mainly glucose(Glu), galactose(Gal) and inositol, and their molar ratio was 6 ∶ 1 ∶ 2, and with a small quantity of glucosamine. There were the chief linkage Glu, Gal, glucosamine and inositol with β-1→4 glycosidic bond and β-1→3 glycosidic bond in the backbone, and there was Glu with β-1→2 glycosidic bond and β-1→6 glycosidic bond in the branch chain. The results showed that PCE-CGL was possible as insulin chemical mediator similar things to acquire its biological effect by increasing the number of chemical mediator.
