摘 要:淡雅型白酒古井贡酒大曲中的真核微生物群落结构一直未在分子水平上得到解析。采用构建ITS rDNA克隆文库的非培养方法对其中温大曲和高温大曲进行真核微生物群落结构及其多样性研究。所得克隆归为21个分类操作单元(OTU),分属子囊菌门、毛霉亚门的毛霉菌目、担子菌门和未培养菌类。中温大曲和高温大曲的真核微生物种类差异较大。中温大曲中真核微生物相对丰富,其中毛霉亚门的毛霉目占46.15%;子囊菌门的曲霉科占26.92%,发菌科占1.92%,未定散囊目占0.96%,酵母目占0.96%;担子菌门占0.96%;未培养类占22.12%。高温大曲的菌种相对比较单一,优势菌为子囊菌门中的嗜热子囊菌科和发菌科,分别占34.62%,58.65%,未培养类占6.73%。另存在一些潜在新种值得进一步研究。Molecular analysis of eukaryotic microbial community structure of Gujinggong liquor has not been reported so far. ITSrDNA clone library technique was conducted to analyse the eukaryotic bacterial community diversity in high-temperature Daqu andmedium-temperature Daqu of Gujinggong liquor. All clone strains were classified to 21 OTUs, belonging to Ascomycota,Mucoromycotina (Mucomles), Basidiomycota and uncultured bacteria. Overall, medium-temperature Daqu eukaryotic bacterialdiversity was significantly more complex than that of high-temperature Daqu. Thermoascaceae (34.62%), Trichocomaceae (58.65%)and uncultures species (6.73%) accounts for the majority of the high-temperature Daqu swains, while both occupy little (1.92%) in themedium-temperature Daqu samples, in which, Mucoromycotina (Mucorales), Ascomycota (Aspergillaceae), Unclassified Eurotiales,Saccharomycetales, Basidiomycota and uncultures species account for 46.15%, 26.92%, 0.96%, 0.96%, 0.96%, and 22.12%,respectively. There are some potential new eukaryotic species that need to be further identified.
