摘 要:通过在60℃,pH值7.0条件下采用3种不同的方法,即QB/T 1803—1993测定法、3,5-二硝基水杨酸比色法(DNS法)、Phadebas amylase test法检测中温α-淀粉酶活力,分析用于检测中温α-淀粉酶活力快捷、方便、准确的方法,从而为其广泛的应用及测定提供依据。结果表明,QB/T 1803—1993测定法准确性高、精密度好,且操作简便、测定快捷,适于测定中温α-淀粉酶活力;3,5-二硝基水杨酸比色(DNS法)测得酶活力范围小,适于定性分析;同样的稀释度Phadebas amylase test法在37,60℃下的测定结果基本一致,而又由于中温α-淀粉酶的最适反应温度在60℃左右,说明该法不适于测定中温α-淀粉酶活力。Under the conditions of 60 ℃, pH 7.0 by using three different methods, including QB/T 1803-1993 assay, 3,5-two nitro salicylic acid colorimetric method (DNS method) , Phadebas amylase test, to detect the activity of medium temperature α-amylase. This assay aims at analysis the most convenient, fast, accurate method and provide the basis for its extensive application and determination. The experimental results show that: the QB/T 1803-1993 method which is not only with high precision and good accuracy, but operates simple and determinates fast is fit for the determination of medium temperature α-amylase activity; 3,5-two nitro salicylic acid colorimetry (DNS) which can determinate small range of enzyme activity is fit for qualitative analysis. With the same dilution degrees under 37 ℃ and 60 ℃ the measurement results of Phadebas amylase test method are basically identical, while the optimum reaction temperature of medium temperature α-amylase is 60 ℃, so this method is not suitable for the determination of medium temperature α-amylase.
