摘  要:确定荞麦苗中抗氧化物质提取的最佳乙醇体积分数,并对荞麦萌发过程中抗氧化活性的变化进行评价。用不同浓度的乙醇提取不同萌发时间荞麦苗中的抗氧化成分,通过DPPH自由基清除能力检测其抗氧化活性并计算提取物的IC50值。结果体积分数60%乙醇溶液提取的得率总体较高,最高值为萌发第10天的25.23%,但提取物的抗氧化活性较低;而体积分数40%乙醇的荞麦苗提取物则显示了较高的DPPH自由基清除能力,随着发芽时间的延长抗氧化能力逐步上升,在萌发第6天的时后到达最高值79.80%,为第3天的1.4倍;综合考虑提取得率与提取物的DPPH自由基清除率,发现在萌发第9天时荞麦苗的抗氧化能力最强。乙醇体积分数对荞麦苗提取物抗氧化能力的变化有一定影响,主要抗氧化物质集中在体积分数40%乙醇中,荞麦萌发过程中抗氧化活性呈增加趋势,第9天时最高。The best concentration of ethanol for the extraction of antioxidant components from buckwheat sprouts was determined, and the changes of antioxidant activity during buckwheat germination were evaluated. Different concentrations of ethanol were used to extract the antioxidant components in the buckwheat sprouts of different germination time. DPPH radical scavenging ability was used to analyze their antioxidant activity and the IC50 value was calculated. 60% ethanol solution had a higher extraction yield, the highest extraction yield was 25.23%, the germination time was 10 d, but antioxidant activity of extracts was low. However, 40% ethanol extracts showed a higher DPPH radical scavenging ability, antioxidant capacity increased when germinated for a longer duration. 6 d germination buckwheat sprouts reached maximum 79.80%, which was 1.4 times for 3 d germinated buckwheat sprouts. After comprehensive consideration of extraction yield and DPPH free radical clearance rate, the buckwheat sprouts germinated for 9 d had the strongest antioxidant capacity. The concentration of ethanol used for the extraction from buckwheat sprouts impacted the antioxidant activity of extract, the main antioxidant components concentrated in 40% ethanol extract, the antioxidant activity of buckwheat increased during the germination period and reached the maximum in the ninth day.
