摘  要:[目的]研究影响酶法回收南极磷虾加工下脚料中的蛋白质及虾青素的因素,以期在提取虾壳中蛋白质的同时又能将虾青素回收,增强蛋白质的应用价值。[方法]以南极磷虾加工下脚料为原料,采用酶解法提取虾青素与蛋白质,考察在碱性蛋白酶作用下不同加热温度、酶底比、加热时间、p H对蛋白质和虾青素回收的影响。[结果]经试验确定,以Alcalase碱性蛋白酶为提取剂,酶解时间2 h,酶底比为3 750 U/g,p H 8.5,酶解温度为50℃时,蛋白质的提取率最高;酶解时间为2 h,酶底比3 750 U/g,p H 8.5,酶解温度为55℃时,虾青素的提取率最高。[结论]研究可为南极磷虾加工下脚料的综合开发利用提供参考依据。[Objective] To study influencing factors for recovery of protein and astaxanthin from Antarctic krill shell,the aim is to strengthen protein application value. [Method] With Antarctic krill shell as raw material,enzymatic hydrolysis was adopted to extract astaxanthin and protein,effects of different heating temperature,enzyme dosage,heating time,p H on protein and astaxanthin were investigated. [Result]Confirmed through experiments,the optimal process parameters for enzymatic hydrolysis to extract protein are: alkaline protease Alcalase as extracting solvent,hydrolysis time 2 h,the amount of enzyme 3 750 U / g,p H 8. 5,hydrolysis temperature 50 ℃. Enzymatic extraction of astaxanthin optimal process parameters are: alkaline protease Alcalase as extracting solvent,reaction time 2 h,the amount of enzyme 3 750 U / g,p H 8. 5,reaction temperature 55 ℃. [Conclusion]The study can provide reference basis for comprehensive development and utilization of Antarctic krill shell.
