摘 要:以自行研制的脉冲磁场杀菌设备对格氏李斯特菌进行杀菌实验,研究了格氏李斯特菌的生长时期、脉冲磁场参数(强度、脉冲数),介质参数(温度、Na+浓度、p H)等对脉冲磁场杀菌效果的影响。结果表明:格氏李斯特菌在对数生长中期对脉冲磁场最为敏感;脉冲磁场对格氏李斯特菌的杀菌效果产生生物学窗口效应,不同的磁场强度杀菌时,2.5T和4.0T残留率出现谷值;不同的脉冲数杀菌时,25个脉冲数残留率达到谷值;当介质温度为30℃时,残留率最低,较低或较高温度的介质中,杀菌效果较差;Na+对脉冲磁场的杀菌效果有一定的协同作用;较低或者较高的p H促进了脉冲磁场的杀菌作用,p H为9.0时残留率最低;对2.5T,25个脉冲数处理的格氏李斯特菌进行扫描电镜观察时发现菌体细胞膜破裂,胞内物质外溢,这可能是脉冲磁场造成格氏李斯特菌死亡的原因。Effect of different growth phase, pulsed magnetic field (PMF)parameters (intensity, pulsed number), medium parameters (temperature, concentration of Na+ , pH) on sterilizing effect of Listeria Grayi by pulsed magnetic field were studied.The results indicated that Listeria Grayi was sensitive to pulsed magnetic field at the middle of log phase.Pulsed magnetic field generated biological window effect on sterilization effect of L.GrayLWhen different intensity used to sterilization,the survival rate reached valley value at 2.5T and 4T.When different pulsed number used to sterilization, the survival rate reached valley value at the number of 25. Low and higher temperature,the sterilization effect was worse, only in 30~C the survival rate reached the minimum value.Na~ had the synergistic effect to sterilization effect.Low and higher pH value increased the sterilizing effect, survival rate reached the minimum value when pH was 9.0. By scanning electron microscope to observe the samples after handled at 2.5T intensity and 25 pulsed number,the result showed that cells membrane rupture and intracellular material spill,this may be the death reason of L.Grayi by PMF.
