摘 要:以30,100和200年窖龄窖泥为样品,采用PCR-DGGE技术探索不同窖龄窖泥细菌类群及群落演变规律.结果表明:不同窖泥样品DGGE图谱均出现14-23条较清晰的条带,其中共有优势条带为第4,11,19号,优势度均在2%以上.多样性指数在2.21-2.58之间,且随窖龄的增加而呈现增加趋势;相似性指数在0.427-0.629之间.30年与200年窖泥细菌群落聚为一类,相似性指数最高,达0.629.30年与100年窖泥细菌群落相似性指数最低,为0.427.PCR-DGGE图谱优势条带割胶测序结果显示:它们属于煎盘梭菌、耐乳酸杆菌、梭菌目细菌、Uncultured bacterium、丁酸梭菌、瘤胃球菌属、酪丁酸梭菌、醋酸杆菌属.Samples of pit mud with a cellar age of 30, 100 and 200 years were analyzed with PCR-DGGE (polymerase chain reaction-denaturing gradient gel them and their succession. Fourteen to 23 distinct eleetrophoresis) to investigate the bacteria clusters in DGGE bands were detected in the samples, of which Bands 4, 11 and 19 were dominant, with a dominance degree of 〉2%. The diversity index tended to increase with increasing cellar age, varying between 2.21 and 2.58. Their similarity indexes ranged between 0. 427 and 0. 629. The bacteria communities in 30-year-old pit mud and in 200-year-old pit mud were clustered into one group, for they had the highest similarity index (0. 629). The bacteria communities in 30- year-old pit mud and in 100-year-old pit mud had the lowest similarity index (0. 427). The PCR-DGGE advantages bands tapping results showed that they belonged to Clostridium sartagoforme, Lactobacillus acetotolerans, Clostridiales bacterium, Clostridium butyricum, Ruminococcus sp. , Clostridium tyrobutyricure and Eubacteriaceae bacterium.
