摘  要:目的 探讨对四氧嘧啶(ALX)模型小鼠具有显著降糖效果的茶多糖组成在小鼠体内的分布规律。方法 依次采用水提醇沉、脱蛋白、酒精分级的方法制备分子量不同的四种茶多糖组分(TPF30,TPF50,TPF70和TPF90),采用GC检测四种组成的单糖组成,考察四种组分对对ALX模型小鼠的降血糖效果,并采用荧光标记的方法检测降糖效果显著的TPF70在小鼠消化道各段及各组织器官中24 h的动态分布规律。结果 酒精分级获得的四种茶多糖组分的得率比为1:13.6:5.5:2.8,四种组分的单糖组成比不同,且其降糖效果也存在差异。降糖效果显著的TPF70在小鼠消化道各段的吸收高峰为1-3 h,随后逐渐下降,并有部分TPF70没被吸收随粪便排除体外。TPF70在内脏器官中的浓度远高于在消化道中的浓度,其中在肝脏中浓度最高,3h达到峰值。结论 本实验为粗老茶叶的开发利用提供了基础研究数据。Objective To investigate the distribution of tea polysaccharide in alloxan (ALX) induced diabetic mice. MethodsThe polysaccharides were extracted from coarse tea leaves and four tea polysaccharide fractions with different molecular weight were obtained (TPF30, TPF50, TPF70 and TPF90). The monosaccharide composition of the four components were detected by GC, and their hypoglycemic effect were evaluated in diabetic mice. The dynamic distributions of TPF70 were monitored in digestive tract and other tissues using fluorescence labeling within 24 h.ResultsThe yield ratio of four components was 1:13.6:5.5:2.8. The monosaccharide composition ratio of the four components were different. There were also differences in the hypoglycemic effect of the four components. The absorption peak of TPF70 in digestive tract was at 1-3 h after administration, and then gradually declined.Some of the TPF70 was not absorbed and discharged from intestine. The concentration of TPF70 in the visceral organs was significantly higher than the concentration in the digestive tract, and the highest concentration was in the liver.ConclusionThis experiment provides the basic data of the polysaccharides from coarse tea leaves for future application.
