摘  要:以氧化铝作层析柱填料,对类球红细菌类胡萝卜素进行分离纯化,采用石油醚-丙酮(9∶1,V/V)、石油醚-丙酮(7∶3,V/V)分别对样品进行洗脱,得到色素提取液Ⅰ和色素提取液Ⅱ。采用高效液相色谱-电子喷雾离子源质谱法(HPLC-ESI-MS/MS)分析类球红细菌类胡萝卜素的组成,其中4个色谱峰得到显著分离,初步鉴定这4个色谱峰对应的组分均属于类胡萝卜素。其中,1号色谱峰和3号色谱峰为同一种组分,即扇贝醇(Pectenol)B,相对分子质量583.4;2号色谱峰和4号色谱峰对应组分依次为葫芦黄素(Cucurbitaxanthin)B,玫红品(Rhodopin),相对分子质量依次为601.3,544.2。In this paper, the carotenoids from Rhodobacter sphaeroides is separated and purified with a chromato-graphic column (1×20 cm) packed with alumina, and the pigment extraction I is obtained by eluting with petroleum ether-acetone (9:1,V/V), the pigment extraction 1/ is obtained by eluting with petroleum ether-acetone(7:3,V/V). Four chromatographic peaks of carotenoids from Rhodobacter sphaeroides were resolved well. And three main compositions of carotenoids from Rhodobacter sphaeroides, which belong to carotenoids, were isolated and identified using high perfor- mance liquid chromatography (HPLC) equipped with a C30 column and electrospray ionization tandem mass spectrometry/ mass spectroscopy (ESI-MS/MS). The fraction which were the main compositions of the four chromatographic peaks prod- ucts, were identified as carotenoids with the help of high performance liquid chromatography-electrospray ionization tan- dem mass spectroscopy/mass spectroscopy (HPLC-ESI-MS/MS). The main compositions of the first and third chromato- graphic peaks products were the same one, it was Pectenol B, and its relative molecular weight was 583.4. The main compositions of the second and fourth chromatographic peaks products were Cucurbitaxanthin B and Rhodopin, and their relative molecular weight were 601.3 and 554.2.
