摘 要:本文探究了浸提条件对聚酯型儿茶素浸出量的影响。在单因素试验的基础上,以液料比、浸提试剂体积分数、浸提温度作为因素,通过三元二次通用旋转组合设计试验优化聚酯型儿茶素的浸提条件,得到聚酯型儿茶素浸出量与浸提条件的回归模型。结果表明:浸提试剂浓度、浸提温度对聚酯型儿茶素浸出量的影响差异极显著,料液比对其影响差异显著;主因素效应为浸提试剂体积分数>浸提温度>液料比。同时确定了最佳浸提条件:48%甲醇,70℃,29.41∶1 m L/g,10 min。在此条件下,聚酯型儿茶素的浸出量为11.63 mg/g,与模型预测值11.70 mg/g接近。In order to study the of extraction conditions on theasinensins extraction in tea, several important factors such as extraction time, extraction temperature, liquid-solid ratio and extraction solvent concentration were studied by one factor at-a-time. By keeping extraction time 10 min, liquid-solid ratio(10∶1-40∶1 mL/g), extraction solvent concentration(40%-70%) and extraction temperature(30-70 ℃) was chosen as the factors of three-factor second order general rotation combination experiment. A model was obtained by regression analysis, we subtracted the terms which were not significant by stepwise regression analysis, then we obtained a simplified model of the relation between the theasinensins extraction and the extraction conditions. The experiment results indicated: the extraction solvent concentration, extraction temperature had extremely significant effect on theasinensins extraction in tea, respectively; the liquid-solid ratio had significant effect on theasinensins extraction in tea; the influencing sequence from significant to common is extraction solvent concentration, extraction temperature, liquid-solid ratio. The optimum extraction parameters were as follows: extraction temperature 70 ℃, liquid-solid ratio 29.41 ∶1 mL/g, extraction solvent concentration 48% methanol, extraction time10 min. By substituting levels of the factors into the quadratic regression equation, the maximum predictable response for theasinensins extraction was calculated and was experimentally verified. The maximum theasinensins extraction obtained experimentally using the optimized extraction conditions was 11.63 mg/g, which was in correlation with the predicted value of 11.70 mg·g^-1by the response surface methodology regression study.
