摘 要:比较罗耳阿太菌胞外多糖(Athelia rolfsii exopolysaccharides,AEPS)提取过程中蛋白脱除方法,并研究其体外抗氧化活性。以除蛋白率和多糖损失率为指标,对比Sevag法、TCA法、等电点法脱蛋白效果,确定最优脱除蛋白方法。以AEPS对Fe^(2+)的螯合能力和还原力,以及对DPPH、羟基自由基的清除能力4个抗氧化指标评估AEPS体外抗氧化活性。结果表明,AEPS最佳除蛋白方法为等电点法。调节发酵液p H为3.5,酸沉时间2 h,蛋白清除率为91.3%,多糖损失率仅为7.2%。与清除·OH相比,AEPS对DPPH具有较强清除效果,且浓度2.5 mg/m L时,对Fe^(2+)的螯合率为74.4%。The methods of deproteinization from fermentation broth of Athelia rolfsii were compared and antioxidant activities in vitro of exopolysaccharides from Athelia rolfsii were studied.Using protein clearance rate and polysaccharide loss rate as indicators,Sevag method,TCA method and isoelectric point method were compared and the best method of deproteinization was determined.Four indicators was used to estimate AEPS antioxidant activity including Fe2+ chelating ability,reducing power and removal abilities of DPPH and ·OH.The data showed that isoelectric point method was the best method of deproteinization for AEPS.Fermentation broth p H3.5 was stay for 2 h,Protein clearance rate reach 91.3% and polysaccharide loss rate was 7.2%.Compared with· OH,AEPS had a strong scavenging effect on DPPH,and chelating rate of Fe2+ was 74.4% when the concentration of AEPS was 2.5 mg/m L.
