摘  要:采用二水平多因素实验设计对影响木醋杆菌和醋酸杆菌混菌发酵细菌纤维素和醋酸的培养基成分进行了筛选,所选取的7个相关因素为糖源、酵母膏、蛋白胨、KH2PO4、MgSO4、无水乙醇、柠檬酸,在此基础上,采用响应曲面法(RSM)对影响木醋杆菌和醋酸杆菌混菌发酵细菌纤维素和醋酸培养基的关键影响因素:糖源、无水乙醇的最佳水平范围做了进一步研究与探讨,通过对二次多项式回归方程求解得知,在上述自变量分别为糖源4.5%,无水乙醇4.0%时,综合指标(综合指标=醋酸产量+细菌纤维素产量)有最大值5.17。优化的混菌发酵培养基为:糖4.5%,酵母膏0.8%,蛋白胨1.0%,KH2PO40.3%,MgSO40.2%,无水乙醇4.0%。在此条件下测得的综合指标为5.21,比未优化前的4.56提高了14.3%。In this paper, the culture medium composition of bacterial cellulose and acetic acid from Acetobacter zylinum and Acetobacter is screened, seven related factors are sugar, yeast extract, peptone, KH2PO4, MgSO4, ethanol, citric acid. On the basis, the further optimum conditions of key influencing factors such as sugar, ethanol are discussed by response surface method. By polynomial quadratic equations, the overall indicator (overall indicator = acetic acid yield q-bacterial cellulose yield) reaches the maximum value of 5. 17 when the above independent variable is 4. 5%, 4. 0% respectively. Therefore, the optimal mixed fermentation medium is sugar 4.5%, yeast extract 0.8%, peptone 1.0%, KH2PO4 0. 3%, MgSO4 0. 2%, ethanol 4.0%. Under such conditions, the overall indicator is 5.21, which is increased by 14.3% compared with 4.56 before optimization.
