摘 要:以斑点叉尾(Ietalurus punetaus)鱼皮为原料,采用响应面法对胶原蛋白热水法提取工艺进行优化。在单因素试验的基础上,使用Design-Expert软件对提取温度、提取p H与料液比进行三因素响应面试验,以胶原蛋白得率为响应值优化热水法提取条件,并验证响应面预测值与实测值的一致性。胶原蛋白最佳提取工艺条件为提取温度100℃、提取p H为6、料液比1∶10(g∶m L),胶原蛋白得率为16.98%。实测值与响应面预测值拟合良好,说明通过响应面试验设计对热水法提取胶原蛋白的优化是有效的。Response surface methodology was applied to optimize the extraction conditions of collagen by heating in water in fish skin of channel catfish (letalurus punetaus). On the basis of single factor tests, response surface analysis was designed by Design-Expert and the effects of extraction temperature ,extraction hydrogen ion concentration and extraction ratio of material to water On the yield of collagen were investigated,and the predicted value and measured value were also contrasted. The results showed the optimal extraction conditions as follows: the extraction temperature was 100 ℃ ,extraction hydrogen ion concentration was 6,extraction ratio of material to water was 1:10 (g:mL). Under these conditions,the yield of collagen was 16.98%,which was consistent with the predicted value. It showed that the experiment was effective.
