摘  要:以酿酒葡萄皮渣为原料,研究并优化双水相法对花色苷得率的影响。在单因素的基础上,通过PlackettBurman实验筛选出p H、乙醇质量分数、硫酸铵质量分数、料液比为主要自变量,以花色苷得率为响应值,利用BoxBenhnken中心组合设计原理和响应面分析法,研究各自变量的交互作用对花色苷得率的影响,模拟得到二次多项式回归方程的模型。响应面优化后的最佳工艺参数是乙醇质量分数为28%,硫酸铵质量分数为18.14%,p H为3.00,料液比为1∶35 g/m L,花色苷得率为4.43 mg/g。此研究将为花色苷实际生产应用提供理论依据。Grape skin was used as the raw material to explore the effect of Aqueous two-phase system on yield ofanthocyanin.Four extraction parameters including pH,ethanol concentration,ammonium concentration,solid-liquidratio were optimized by using Plackett-Burman design based on single factor investigations for achieving yield ofanthocyanin.The interaction of the respective variables and their influence on the yield were studied by using Box-Benhnken central composite design and response surface analysis theory, the simulated quadratic polynomialregression equation of prediction model was set up.The optimal parameters were optimized by response surfaceanalysis.The ethanol concentration, ammonium sulphate concentration, pH and solid-liquid ratio were 28%,18.14% ,3.00 and 1 35 g/mL, respectively.The optimal yield of anthocyanin was 4.43 mg/g.The research will providetheoretical support for the anthocyanin of practical application.
