摘  要:目的:提高柑橘膳食纤维(DF)中水溶性膳食纤维含量,获得功效均衡的膳食纤维产品。方法:从自然发霉的柑橘果实表面分离得到1株纤维素酶活力较高的菌株CIs16,根据其形态学特征及18S r DNA序列分析结果进行鉴定。对CIs16发酵的柑橘皮渣进行主要成分及性能分析,测定其中水溶性膳食纤维(SDF)、总膳食纤维(TDF)含量、SDF/TDF比例、产品持水力与溶胀性。结果 :CIs16菌落呈短绒状,孢子暗绿色,具有帚状分生孢子,结合其18S r DNA序列分析结果,鉴定为青霉属菌株。CIs16在柑橘皮渣培养基中生长良好,28℃发酵3 d所得产物中SDF、TDF产率及SDF/TDF比例分别由发酵前的7.61%,59.27%,12.45%上升至23.52%,83.24%,27.62%,持水力由5.85 g/g下降为4.41 g/g,溶胀性由6.92 m L/g增加至9.58 m L/g。结论 :以CIs16为菌株发酵柑橘皮渣,能够显著提高产品中SDF含量,为高品质DF的制备及柑橘皮渣的综合利用提供理论和技术基础。Objective: To improve the content of soluble dietary fiber in total dietary fiber, and obtain function-balanced DF product. Method: A filamentous fungi of high cellulase activity, named CIs16, was isolated from moldy citrus fruit and identified through morphologic characters and 18 S rDNA sequence analysis. Citrus dregs fermented by CIs16 for three days under 28 ℃, and the main components, SDF content, TDF content, SDF/TDF ratio, water-holding capacity and swelling property of the product were investigated. Results: CIs16 with floss-shaped colony, dark green spores and brush-like conidiophore, was identified as Penicillium sp. through its morphologic characters and 18 S rDNA sequence analysis. SDF content, TDF content and SDF/TDF ratio in fermented citrus dregs was increased from 7.61%, 59.27%%and 12.45% to 23.52%, 83.24% and 27.62% respectively. The water-holding capacity of fermented dietary fiber was decreased from 5.85 g/g to 4.41 g/g and swelling property increased from 6.92 mL/g to 9.58 mL/g. Conclusions: CIs16 grew well in citrus dregs medium and its SDF content in fermented samples was improved significantly. The experimental results provided theoretical and technological basis for producing dietary fiber of high quality and comprehensive utilization of citrus dregs.
