摘 要:为确定一种快速简便、准确可靠的酵母菌油脂含量测定方法,对磷酸香草醛显色法、氧化酶法与重量法进行比较分析。结果表明:在测定酵母菌体总脂含量时重量法准确度和精确度高,成本低,但操作过程烦琐、耗时长,溶剂有毒易污染环境;磷酸香草醛显色法的平均回收率约为93%,准确度和精确度较高,耗时少,最低检测限约为0.08 g/L,测定时需与相应的菌体油脂标准曲线匹配使用,其测定发酵液油脂含量与重量法差异不显著(P>0.05),可以作为酵母菌油脂含量测定的方法;在测定酵母菌体内甘油三酯含量时,氧化酶法平均回收率达到98.0%,准确度和精确度高,该方法操作简便,耗时少,最低检测限为1.2 g/L;氧化酶法需与甘油三酯标准曲线匹配使用,同时还需对细胞进行破壁,但由于其快速准确性,也不失为一种酵母菌油脂含量测定的好方法。In order to find out a rapid, convenient and reliable determination method of lipid content in yeast, weight method, sulfo -phospho -vanillin reaction method and oxidase method were compared. The results showed that when determining the total lipid content in yeast, the weight method had the items of high accuracy and precision, low cost, complicated operation, more time - consuming, toxic solvents and environmental pollution. The average recovery rate and limit of detection of the sulfo - phospho - vanillin reaction method were about 93% and 0.08 g/L respectively. The sulfo - phospho - vanillin reaction method had the items of high accuracy and precision, and less time -consuming. Though the results had to be matched with the corresponding standard curve of lipid content, this method had no significant differ- ence in lipid content compared with weight method( P 〉 0.05 ) ,so it could be used to determine the lipid content in yeast. When determining the triglyceride content in yeast, average recovery rate and limit of de- tection of the oxidase method were 98.0% and 1.2 g/L respectively. The oxidase method had the items of high accuracy and precision, easy operation and less time - consuming. Though the results had to be matched with the standard curve of triglyceride content and the cells should be disrupted, oxidase method was a good method for the determination of lipid content in yeast because of its rapidity and preci- sion.
