摘  要:以ACE抑制率为指标,比较了各种蛋白酶对青柳蛤的水解效果。在单因素试验的基础上,通过响应面试验设计对酶解条件进行了优化,试验结果表明:碱性蛋白酶对青柳蛤蛋白的水解效果由于其他蛋白酶,该酶的最优水解条件为:pH 8.0,酶的添加量2 632.4 U/g,水解时间2.3 h,温度50℃。经优化,青柳蛤蛋白水解液的ACE抑制率达到52%。水解液经干燥后灌胃SHR大鼠,体内试验表明青柳蛤酶解物具有体内辅助降血压活性,血压降幅在3 947 Pa左右。Using ACE inhibition rate as the index, the effects of several proteases were compared. On the basis of single factor experiment and the response surface design of experiment, response surface optimization had been done. The results indicated that alkali protease was the best of all. The optimal levels were obtained, including pH 8.0, the ratio of enzyme to substrate of 2 632.4 U/g, 2.3 h, temperature of 50 ℃. After optimation, the ACE inhibition rate increased to 52%. After drying, the hydrolysate was prepared for SHR by oral administration. The results suggested that the hydrolysates of Mactra chinenesisshowed auxiliary hypotensive activity in vivo. The average blood pressure of SHR could go down about 3 947 Pa.
