摘 要:从野生木蹄层孔菌子实体中分离得到药用真菌木蹄层孔菌。基于菌丝体生物量和胞外多糖的含量,通过正交优化确定了木蹄层孔菌深层培养的培养基及培养条件。结果表明,最适培养基配方为蔗糖10.0 g/L、蛋白胨5.0 g/L、CaCl_2 1.0 g/L、MgSO_4 0.25 g/L、V_(B2) 0.5 mg/L,培养条件为25℃,初始pH6.5,摇床转速150 r/min,培养时间为7 d。在此条件下,木蹄层孔菌多糖的产量为3.52g/L,比未优化前提高了4倍,通过检测确定木蹄层孔菌胞外多糖具有一定的体外抗氧化活性。A medicinal mushroom Fomes fomentarius, was isolated from the fruiting body of a wild F. fomentarius. Using orthogonal matrix method, the optimization of submerged culture conditions and nutritional requirements of mycelial biomass and exopolysaccharide(EPS) production from F. fomentarius was studied. The optimal medium for EPS production was that sucrose 10.0 g/L, peptone 5.0 g/L, CaCl2 1.0 g/L, MgSO4 0.25 g/L and V(B2) 0.5 mg/L. The suggested culture conditions were 25 ℃, initial pH 6.5 with a rotary speed of 150 r/min for 7 d. The yield of EPS was 3.52 g/L, which was about four times higher than that at the basal medium. The anti-oxidative activities of EPS in Fomes fomentarius were also characterized using various in vitro assay.
