酒酒球菌磷酸-β-葡萄糖苷酶基因的克隆及生物信息学分析

摘　　要：酒酒球菌是与葡萄酒相关的乳酸菌,具有糖苷酶活性,能够通过水解葡萄衍生的芳香化合物前体以增强葡萄酒香气。采用PCR法从酒酒球菌基因组DNA中克隆得到一个磷酸-β-葡萄糖苷酶基因。序列分析显示:该基因包含一个完整的开放阅读框,全长1 443 bp,编码480个氨基酸残基。其编码的氨基酸序列与酒酒球菌PSU-1的磷酸-β-葡萄糖苷酶序列同源性在99%以上。生物信息学分析显示,该基因编码的蛋白分子质量为55 164.4 u;理论等电点为6.14,疏水性较弱;无信号肽,无跨膜区,为可溶性酶,主要存在于细胞质内,属于糖苷酶家族1,bglB超家族。对酒酒球菌bgl-2基因的克隆与分析,为进一步探究β-葡萄糖苷在酒酒球菌细胞内的代谢研究奠定了理论基础。Oenococcus oeni,as wine-related lactic acid bacteria,shows glycosidase activities,positively contributing to wine aroma by the hydrolysis of grape-derived aroma precursors.In this study,the phospho-β-glucosidase gene was successfully cloned by PCR from the genomic DNA of O.oeni.The sequence analysis showed that the phospho-β-glucosidase gene has a complete opening reading frame sequence contains 1 443 bp,encoding 480 amino acids.Polygenetic analysis showed that the sequence is similar to the bgl member in O.oeni PSU-1,with the similar coefficient of 99%.Bioinformatic analysis showed that the encoded protein with molecular mass of 55 164.4 u and a pI of 6.14.The bgl protein was soluble protein and localized in the cytoplasm,belonging to the superfamily of bglB,Glyco_hydro_1.The cloning and analysis of bgl-2 gene in O.oeni are helpful to further explore of β-glucosides metabolism in bacterial cells.
