摘  要:以油莎草醇提物为原料,利用D101大孔树脂,依次用0,30%,50%,70%和95%乙醇对油莎草醇提物进行梯度洗脱,以DPPH·自由基清除率为考察指标,确定抗氧化活性最强的洗脱部位(目标醇提物),薄层层析(TLC)定性分析,利用50目ODS RP-18反相硅胶对目标醇提物进行分离纯化,再经Sephadex LH-20葡聚糖凝胶柱分离得到近似单体化合物。利用核磁共振(NMR)对单体化合物进行结构鉴定。结果表明,30%乙醇洗脱部位的抗氧化活性最强,其IC50为0.050 68±0.092 74 mg·m L^(-1),弱于阳性对照VC(IC50为0.011 92±0.060 mg·m L^(-1))和芦丁(IC50为0.015 42±0.081 4 mg·m L^(-1)),从目标醇提物中分离得到4个单体化合物,化合物1为7-羟基-6-甲氧基-香豆素,化合物2、3为异荭草苷,化合物4为荭草苷。The Cyperus alcohol extract as raw material, use D101 macroporous resin and gradiently elute with 0, 30%, 50%, 70% and 95% ethanol on D101 macroporous resin. Alcohol extract targeted with the strongest antioxidant activity was confirmed by DPPH free radical scavenging ability. Thin-layer chromatography (TCL) was applied in qualitative identification. Using reversed phase silica gel column (ODS RP-18 reversed phase silica gel, 50 mesh) chromatography quickly separated and purified target alcohol extract, monomer compounds were gained through Sephadex LH-20, the structure of monomer compounds were identified by nuclear magnetic resonance (NMR). The results showed that 30% ethanol elution parts existed the strongest antioxidant activity with IC50 0.050 68±0.092 74 mg·mL^-1, which was weaker than that of positive control VC (IC50 0.011 92±0.060 mg·mL^-1) and rutin (IC50 0.015 42±0.081 4mg·mL^-1). Four monomer compounds were isolated from the target alcohol extract, and compound 1 was 7-hydroxyl-6-methoxy radical coumarin, compounds 2, 3 both were isoorientin, and compound 4 was orientin.
