摘 要:在不同条件下对琼胶进行酸水解,得琼胶寡糖A1(以二、四糖为主)和A2(以己糖、辛糖为主)采用化学发光法和DPPH体系分别研究A1、A2对O2·-、·OH和DPPH3种自由基的清除作用。结果表明,A1、A2对3种自由基都有较好的清除作用,而且清除活性随糖浓度的增加而加强。在O2·-体系中,A2的IC50为0.54mg/mL,效果要好于A1(IC50为0.8mg/mL)。在·OH体系中,A2对·OH的清除作用比A1效果好,IC50分别为1.2和2.3mg/mL。2个体系比较发现A1、A2对O2·-的清除作用高于对·OH的清除作用。在DPPH体系中,A1、A2对DPPH有一定的清除,但A1较A2效果好,IC50分别为4和6 4mg/mL。由此可见,琼胶寡糖在不同体系中清除自由基的活性不同。琼胶寡糖在抗氧化方面具有很好的应用价值。<Abstrcat> The agar oligosaccharides A1(mainly with disaccharidae and tetrasaccharide) and A2 (mainly with hexose and octoses) were prepared from agar under different acid hydrolysis conditions. Using the methods of Luminesecence and DPPH system, the scavenging effects of A1 and A2 on O2·-,·OH and DPPH are determined that A1 and A2 have strong scavenging effects on O2·- with the IC50 at 0.8 and 0.54 mg/mL. The scavenging activities of A1 and A2 on ·OH are lower than those on O2·- and their IC50 on ·OH are 2.3 and 1.2 mg/mL. A2 has better scavenging ability on O2·-and ·OH than A1. A1 and A2 have certain scavenging activities on DPPH and A1 shows higher scavenging ability than A2, which is quite different from the results in O2·- and · OH. But their abilities are much lower than probucol and Vc, which have very active hydrogens and good scavenging activity on DPPH. It suggests that agar oligosaccharides A1 and A2 have different scavenging abilities in different free radical assays.
