摘 要:建立水浴振荡提取-分光光度法测定黑米中花青素含量的方法。考察检测波长和建立标准曲线;应用响应面优化试验确定了水浴振荡提取黑米中花青素的最优提取条件。试验得水浴振荡提取的最优提取条件:溶剂是含0.8%盐酸的50%乙醇水溶液、液固比20∶1(mL∶g)、提取次数2次、温度75℃、时间20 min,花青素得率为1.249%。结果表明水浴振荡提取-分光光度法测定黑米中花青素含量的方法操作简便、重复性好、测定结果精密度高。Spectrophotometry detection method of anthocyanin in black rice with water bath shaking extraction was established. The detection wave length and standard curve were studied, and the optimum extraction conditions of anthocyanin were obtained through response surface design. The optimum extraction conditions were as follows: solvent of 50 % ethanol aqueous solution containing 0.8 % hydrochloric acid, the liquid-solid ratio of 20 ∶ 1(mL/g), extraction times of two times, extraction temperature of 75 ℃,extraction time of 20 min. The extraction yield of anthocyanin was 1.249 %. The results showed that the water bath shaking extraction method and spectrophotometry detection method of anthocyanin in black rice had the advantages of simple operation, good repeatability and high precision.
