摘 要:本文研究植物乳杆菌发酵大麦β-葡聚糖(LFBG)和未发酵大麦β-葡聚糖(RBG)对秀丽隐杆线虫脂肪沉积的影响。在建立秀丽线虫肥胖模型的基础上,分别以不同浓度的LFBG和RBG处理线虫,通过油红O染色和荧光定量PCR法分析线虫体内脂肪含量变化及可能机制。LFBG和RBG中的β-葡聚糖含量分别达到71.70%和72.60%,其重均分子量(Mw)分别为3.14×10~4和1.66×10~5。LFBG和RBG均能显著提高线虫的运动行为能力,从而增加线虫的能量消耗,以减少线虫体内脂肪沉积,且在相同剂量时LFBG的抑制效果优于RBG。当浓度为25μg/mL、50μg/mL和100μg/mL时,与模型组相比,LFBG组的脂肪含量分别减少了15.83%、22.32%和35.67%,RBG组线虫在浓度为25μg/mL时无显著性变化,在浓度为50μg/mL,100μg/mL时脂肪含量分别减少了16.28%和22.29%。此外,LFBG能显著下调daf-16 mRNA的表达量,而RBG可上调cpt-1 mRNA的表达量。由此可见,LFBG和RBG可能通过不同作用途径来抑制线虫的脂肪沉积。In this work,the effects of the Lactobacillus plantarum dy-1 in fermented barley β-glucan(LFBG)and raw barley β-glucan(RBG)on the fat deposition of Caenorhabditis elegans(C.elegans)were studied.The C.elegans,as the model organism,was treated with different concentrations of LFBG and RBG.The changes of fat content and possible mechanism of C.elegans were analyzed by oil red O staining and real-time PCR,respectively.The β-glucan content in LFBG and RBG reached 71.70% and 72.60%,respectively,and the molecular weight(Mw)of LFBG and RBG were 3.14×10^4 and 1.66×10^5,respectively.Compared with the model group,LFBG and RBG could significantly improve the movement behavior of C.elegans,thereby increasing the energy consumption of the nematode to reduce the fat accumulation of nematodes.The fat contents of LFBG treatment decreased by 15.83%,22.32% and 35.67% on the fat deposition when the concentration was 25μg/mL,50μg/mL and 100μg/mL,respectively.The inhibition effect of LFBG was more significant than RBG,which showed no effect when the concentration was 25μg/mL.The fat contents of RBG treatment decreased by 16.28% and 22.29% when the concentration reached 50μg/mL and 100μg/mL,respectively.LFBG could significantly down-regulate the expression of daf-16 mRNA,while RBG could up-regulate the expression of cpt-1 mRNA.In conclusion,LFBG and RBG could effectively regulate the accumulation of fat in C.elegans,through different pathways in C.elegans.
