Optimization of fermentation conditions of cellulase-producing strains from Tibetan cattle by response surface methodology

摘  要:在单因素试验基础上,采用Plackett-Burman试验得出麸皮添加量、蛋白胨添加量及发酵温度是影响菌株产酶的3个最重要的因素。利用Box-Behnken试验设计对类芽孢杆菌(Paenibacillus)N30发酵生产纤维素酶的产酶条件进行响应面优化。结果表明,单因素试验优化结果为麸皮添加量2.5%、蛋白胨添加量0.5%、初始pH值为7.0、发酵温度37 ℃、接种量1.5%、转速150 r/min、装液量125 mL/250 mL、种龄32 h、KH2PO4 0.2%;响应面优化后发酵条件为麸皮添加量2.8%,蛋白胨添加量0.5%,发酵温度38 ℃。在此优化条件下,纤维素酶酶活为42.5 U/mL,是优化前酶活(12.1 U/mL)的3.5倍。On the basis of single factor experiments, the results showed that bran addition, peptone addition and fermentation temperature were the three most important factors affecting cellulase production by Plackett-Burman tests. The cellulase production conditions of Paenibacillus N30 were optimized by response surface methodology. The results of single factor optimization were bran 2.5%, peptone 0.5%, initial pH value 7.0, fermentation temperature 37 ℃, inoculum 1.5%, rotational speed 150 r/min, liquid loading volume 125 ml/250 ml, seed age 32 h, KH2PO4 0.2%. The fermentation conditions optimized by response surface methodology were obtained as follows: bran 2.8%, peptone 0.5% and fermentation temperature 38 ℃. Under these optimal conditions, the cellulase activity was 42.5 U/ml, which was 3.5 times that of before optimization (12.1 U/ml).
