摘 要:目的建立高效液相色谱法同时测定奶味饮料中的香兰素和乙基香兰素的检测方法。方法样品经含5%乙酸的乙腈溶液提取液提取,超声处理后,使用乙腈定容,高速离心后上清液过0.22μm微孔滤膜。使用高效液相色谱检测,检测波长308nm,外标法定量。结果本方法前处理简便易操作,香兰素和乙基香兰素在0.1~100μg/mL浓度范围内线性关系良好,相关系数均大于0.999,以3 S/N作为最低检出限,方法的检出限均为0.02μg/mL,加标回收率分别为97.0%~101.1%、96.0%~100.5%,相对标准偏差小于5%。结论该方法操作简便,准确度高,重现性好,适合奶味饮料中的香兰素和乙基香兰素的快速测定。Objective To establish a simultaneous determination of vanillin and ethyl-vanillin in milk-taste-drink by high performance liquid chromatography.Methods The samples were extracted by acetonitrile with 5%acetic acid solution,after ultrasonic treatment,acetonitrile was used to fix the volume.,Then determined by high performance liquid chromatography at 308 nm after high speed centrifugation and supernatant treatment.The external standard method was used for quantitative analysis.Results The pretreatment method was simple and easy to operate.The linear relationship between vanillin and ethyl vanillin was good in the concentration range of 0.1–100μg/mL,and the correlation coefficient was greater than 0.999.The detection limit of the method was 0.02 g/mL with 3 S/N as the lowest detection limit.The standard recovery was 97.0%–101.1%and 96.0%–100.5%,respectively,with the relative standard deviation less than 5%.Conclusion The method is simple,accurate and reproducible.It can be applied to the simultaneous determination of vanillin and ethyl-vanillin in milk-taste-drink.
