摘 要:将超高压技术和热水提取法用于黑毛尖(HMJ)和千两茶(QL)活性物质的提取,并测定了各种黑茶提取物对α-葡萄糖苷酶和二肽基肽酶(DPP4)的抑制作用。结果表明,超高压提取法得率为13.57%~14.22%,高于热水提取法的得率(10.45%~12.45%)。对α-葡萄糖苷酶的抑制作用最强的是千两茶的热水提取物(IC50=4.91×10^—5 mg/mL),2 mg/mL千两茶热水提取物的抑制率达98.51%,相同浓度的千两茶超高压提取物的抑制率仅达63.57%。对DPP4抑制作用最强的是黑毛尖的热水提取物(IC50=2.9×10^—1 mg/mL),2 mg/mL黑毛尖热水提取物的抑制率达71.14%,相同浓度的千两茶热水提取物的抑制率仅达59.26%。可见,黑茶提取物有显著的降血糖作用,可用于降血糖保健食品或药品的开发,但黑茶的品种和提取方法对其降血糖活性影响较大。To study influence of extraction methods on hypoglycemic activity of different varieties of dark tea,ultrahigh pressure(UHP)technique and hot water bath were applied to two kinds of dark tea extraction,Heimaojian(HMJ)and Qianliang(QL).The inhibiton activity of dark tea extracts onα-glucosidase and DPP4 were determined.Consequently,UHP technique gained extraction rate of 13.57%~14.22%,higher than hot water bath(10.45%~12.45%).Hot water bath QL extract showed the greatest potency onα-glucosidase inhibiton with an IC50 value of 4.91×10^—5 mg/mL,and 98.51%inhibition rate when present at a concentration of 2 mg/mL,while the inhibition rate for UHP QL extract is 63.57%.Moreover,hot water bath HMJ extract exhibited highest DPP4 inhibition activities with the IC50 values of 2.9×10^—1 mg/mL,whose inhibitory rate was 71.14%at the concentration of 2 mg/mL,while the inhibition rate for hot water bath QL extract is 59.26%.In conclusion,the extraction methods and dark tea varieties have great effect on hypoglycemic activity,and all dark tea extracts displayed inhibition on two emzymes,so they could be good raw material for hypoglycemic agents.
