摘 要:为研究玉米胚粕与玉米黄浆新型培养基发酵生产谷胱甘肽的能力,选取糖化且经过三效浓缩后的玉米胚粕与玉米黄浆为原料,制成新型酵母菌发酵培养基,对酿酒酵母S.cerevisiae L5267进行发酵。通过单因素试验与正交试验选取最优条件。利用琥珀酸二酯磺酸钠与异辛烷形成的反胶束体系对谷胱甘肽进行反胶束萃取,萃取后检测萃取液中谷胱甘肽含量。结果表明,玉米胚粕与玉米黄浆糖化液DE值为92;培养基配方:L-cys 0.4 g、MgSO_(4)0.125 g、肌醇0.0375 g、K^(+)0.2 mol/L、糖化液100 g、蒸馏水150 g。在此条件下,按照接种量7%,发酵温度30℃,发酵时间23 h进行发酵。发酵后经琥珀酸二酯磺酸钠与异辛烷反胶团萃取后,最终谷胱甘肽的含量可达到10.01 mg/g,再经过纳滤膜分离设备与低温冷冻干燥后得到谷胱甘肽固体成品。In order to study the ability of glutathione produced by maize embryo meal and maize yellow pulp culture medium fermentation,saccharified and concentrated maize embryo meal and maize yellow pulp are selected as the raw materials to prepare a new yeast fermentation culture medium for S.cerevisiae L5267 fermentation.The optimal conditions are selected by single factor test and orthogonal test.Glutathione is extracted by the reverse micelle system formed by sodium diethylhexyl sulfosuccinate and isooctane,and the content of glutathione in the extraction solution is detected after extraction.The results show that when the DE value of maize embryo meal and maize yellow pulp is 92,the culture medium ratio is L-cys 0.4 g,MgSO_(4)0.125 g,inositolum 0.0375 g,K^(+)0.2 mol/L,saccharified solution 100 g,distilled water 150 g.Under these conditions,the fermentation is conducted when the inoculation amount is 7%,the fermentation temperature is 30℃and the fermentation time is 23 h.After fermentation,the final glutathione content reaches 10.01 mg/g after extraction with sodium diethylhexyl sulfosuccinate and isooctane reversed micelles.Glutathione solid products are obtained by nanofiltration membrane separation equipment and low-temperature freeze drying.
