摘  要:通过静态吸附-解吸试验从5种大孔树脂筛中筛选出D-101大孔树脂对紫玉米穗轴花青素进行纯化,结果表明:D-101的静态吸附率(94.17±1.31)%,解吸率(90.83±1.33)%,为最佳纯化材料。通过动态吸附-解吸试验确定最佳纯化工艺条件:上柱体积9 BV、上样流速1.5 m L/min、洗脱剂为体积分数70%乙醇与体积分数0.02%盐酸的混合液(体积比100∶1)、洗脱流速1.5 m L/min、洗脱体积8 BV。在此条件下,获得穗轴花青素纯度为29.76%,相比于原料花青素纯度0.74%,提高了约40倍。Through static adsorption desorption test,D-101 macroporous resin was selected from five macroporous resin sieves to purify purple corn cob anthocyanins. The results showed that the static adsorption rate of D-101 was( 94. 17 ± 1. 31) %,the desorption rate was( 90. 83 ± 1. 33) %,and D-101 was the best purification material. The optimal purification process conditions were determined by dynamic adsorption desorption test: the upper column volume was 9 BV, the sample loading flow rate was1. 5 m L/min,the eluent was the mixture of volume fraction70 % ethanol andvolume fraction 0. 02 % hydrochloric acid( volume ratio 100 ∶ 1),the elution flow rate was 1. 5 m L/min,and the elution volume was 8 BV. Under these conditions,the purity of cob anthocyanins obtained was 29. 76 %,which was about 40 times higher than that of raw material anthocyanins of 0. 74 %.
