摘  要:采用响应面法优化铁皮石斛叶多酚的提取工艺,并考察铁皮石斛叶多酚对自由基的清除作用。结果表明:最优铁皮石斛叶多酚提取工艺条件为乙醇体积分数50%,液料比36∶1(m L/g),超声功率350 W,提取时间37 min,提取温度60℃,提取2次。在最优条件下铁皮石斛叶多酚提取量为34.15 mg/g。铁皮石斛叶多酚可以有效清除羟自由基、超氧阴离子自由基及DPPH自由基,铁皮石斛叶多酚自由基清除率随着多酚质量浓度增加而升高,其对羟自由基、超氧阴离子自由基及DPPH自由基的半数清除率(IC50)分别为0.554、0.775 mg/mL及0.596 mg/mL,较同浓度的VC弱。The aim of this study was to optimize the process of extracting polyphenols from Dendrobium officinale leaves using response surface methodology and to investigate the free radical scavenging ability of polyphenols from D. officinale leaves. The following extraction conditions were found to be optimal:ethanol concentration,50%;ultrasonic power,350 W;extraction time,37 min;liquid-solid ratio,36∶1(mL/g);extraction temperature,60 ℃;and times of extractions,2. Under these optimized parameters,the yield of polyphenols from D. officinale leaves was 34.15 mg/g. Moreover,the polyphenols from D. officinale leaves effectively scavenged hydroxyl radicals,superoxide anion radicals,and 2,2-diphenyl-1-picrylhydrazyl(DPPH)free radicals. The free radical removal rate increased with increasing polyphenol concentration. The half-maximal scavenging concentrations(IC50)of the polyphenols toward hydroxyl radicals,superoxide anion radicals,and DPPH free radicals were 0.554,0.775 mg/mL and 0.596 mg/mL,respectively. However,the antioxidant capacity of the polyphenols was weaker than the antioxidant capacity of vitamin C at the same concentration.
