摘 要:以红发夫酵母(Phaffia rhodozyma)PR2为出发菌株,采用紫外(UV)与硫酸二乙酯(DES)进行复合诱变,通过二苯胺平板初筛、摇瓶复筛、遗传稳定性试验,筛选高产虾青素突变菌株,并通过单因素试验对其发酵产虾青素条件进行优化。结果表明,在紫外照射100 s,DES含量1%的最适诱变条件下筛选得到一株高产虾青素突变菌株PUD11,其虾青素产量达到22.16 mg/L,且该突变菌株传代8次仍具有良好的遗传稳定性,其产虾青素的最佳发酵条件为发酵温度22℃、初始pH值5.0、接种量10%、摇瓶转速220 r/min、装液量75 mL/300 mL、发酵时间120 h。在此优化条件下,虾青素产量为31.39 mg/L,较出发菌株提高了106.24%。Using Phaffia rhodozyma PR2 as the starting strain,the strain was mutated by ultraviolet(UV)and diethyl sulfate(DES)compound mutation,and the high-yield astaxanthin mutant strain was screened through diphenylamine plate,shake flask and genetic stability test.The fermentation conditions of the mutant strain for astaxanthin production were optimized by single factor experiments.The results showed that a high-yield astaxanthin mutant strain PUD11 was screened under the optimal mutagenic conditions(UV irradiation for 100 s and DES content 1%),and the astaxanthin yield reached 22.16 mg/L.The mutant strain still had good genetic stability after 8 generations.The optimum fermentation conditions of mutant strain for astaxanthin production were as follows:fermentation temperature 22℃,initial pH 5.0,inoculum 10%,shaking flask speed 220 r/min,liquid volume 75 ml/300 ml and time 120 h.Under the optimized conditions,the astaxanthin yield was 31.39 mg/L,which was 106.24%higher than that of the starting strain.
