摘 要:墨红玫瑰产量高,为国内主要食用玫瑰品种。其中含有大量多酚,但目前对其研究不足。该文以墨红玫瑰为原料提取多酚,通过LC-MS分析玫瑰提取物(rose polyphenols extraction,RPE)组成并评价稳定性,以6种常见抗氧化剂:维生素C、没食子酸、α-生育酚、丁基羟基茴香醚、特丁基对苯二酚、茶多酚和金属离子螯合剂EDTA等为参照,通过自由基清除能力、铁离子还原能力以及金属离子螯合能力等指标评价其抗氧化能力。结果表明,从RPE中鉴定出矢车菊素-3,5-二葡萄糖苷、鞣花酸、儿茶素、槲皮素、杨梅素、山奈酚、bis-六羟基二苯酰基-葡萄糖苷和没食子儿茶素等8种多酚,RPE在pH≤4以及温度≤20℃时较为稳定。RPE在对ABTS阳离子自由基、DPPH自由基清除及铁离子还原实验中均表现出良好的抗氧化性能;相同浓度下,其抗氧化能力强于所选6种常见抗氧化剂;在0.1 mg/mL质量浓度下,RPE对ABTS阳离子自由基(7 mmol/L)和DPPH自由基(0.14 mmol/L)的清除率分别为(89.53±0.09)%和(100±0.00)%;但其金属离子螯合能力不及EDTA。该研究结果表明RPE具有较强的抗氧化能力,有望为其应用提供一定的理论和技术支撑。Red rose(Rosa Crimson Glory)is the main edible rose in China,with the characteristics of high yield and rich in polyphenols,however,the antioxidant properties of polyphenols in it have not been studied enough.In this study,polyphenols were extracted and the stability of rose polyphenols extraction(RPE)was evaluated.The antioxidant capacity of RPE was evaluated using vitamin C,gallic acid,α-tocopherol,butyl hydroxyanisole,terbutyl hydroquinone,tea polyphenol,and EDTA as controls by free radical scavenging ability,metal ion chelating ability,and iron ion reducing ability.Results showed that eight polyphenols were identified from RPE,including cyanidin-3,5-diglucoside,ellagic acid,catechin,quercetin,myricetin,kaempferol,bis-hexahydroxydiphenoyl-glucoside,and gallic catechin,which was stable at pH≤4 and lower than 20℃.The obtained RPE showed good antioxidant activity in scavenging ABTS cationic radicals and DPPH free radicals and in the reduction of iron ions.At the same concentration,the antioxidant activity of RPE was stronger than that of the selected 6 kinds of common antioxidants.At the concentration of 0.1 mg/mL,the scavenging rates of RPE on ABTS cationic radicals(7 mmol/L)and DPPH free radicals(0.14 mmol/L)were(89.53±0.09)%and(100±0.00)%,respectively,but the metal ion chelating ability of RPE was lower than that of EDTA.Results show that RPE has strong antioxidant activity,which is expected to provide theoretical and experimental support for its further application.
