摘 要:以玉米皮渣为原料,采用海鲜菇发酵法制备阿魏酰低聚糖(FOs)。以FOs生成量为指标,先通过单因素试验确定海鲜菇发酵用碳源、氮源及矿质离子,再通过正交试验确定最佳培养基组成,然后再对发酵条件进行优化。结果表明:优化后的碳源为D-半乳糖,氮源为牛肉膏,矿质离子为磷酸二氢钾;最佳培养基组成为D-半乳糖5 g/L、牛肉膏2 g/L、磷酸二氢钾0.1 mol/L,与培养基组成成分优化前相比,海鲜菇的纤维素C1酶酶活提高了44.1%,木聚糖酶酶活性提高了164.0%;最佳发酵条件为装液量125 mL、接种量12%(以培养基体积计)、pH 5.5、发酵温度25℃,与发酵条件优化前相比,海鲜菇的纤维素C1酶酶活略有减小,木糖酶酶活提高了40.3%,淀粉酶酶活提高了25.6%。Feruloyl oligosaccharides(FOs)were prepared from corn bran residue by Hypsizygus marmoreus fermentation method.Taking the production of FOs as the index,the carbon source,nitrogen source and mineral ions for Hypsizygus marmoreus fermentation were determined by single factor experiments.The optimal medium composition was determined by orthogonal test,and then the fermentation conditions were optimized.The results showed that the optimal carbon source was D-galactose,the nitrogen source was beef paste,and the mineral ion was potassium dihydrogen phosphate.The optimal medium group was D-galactose 5 g/L,beef paste 2 g/L,potassium dihydrogen phosphate 0.1 mol/L.Compared with the composition of the culture medium optimized before,the cellulase C1 enzyme activity of Hypsizygus marmoreus increased by 44.1%,and the xylanase activity increased by 164.0%.The optimal fermentation conditions were liquid volume 125 mL,inoculum volume 12%(based on the volume of medium),pH 5.5,fermentation temperature 25℃.Compared with the fermentation conditions before optimization,the cellulose C1 enzyme activity of Hypsizygus marmoreus slightly decreased,the xylanase activity increased by 40.3%,and the amylase enzyme activity increased by 25.6%.
