摘 要:采用酶辅助法提取高粱壳多酚。以多酚含量为指标,采用单因素试验和正交试验先确定最佳酶用量,然后进一步优化酶解条件,并对高粱壳多酚进行结构表征和抗氧化活性分析。结果表明:最佳酶用量(以高粱壳粉质量计)为木瓜蛋白酶2.1 mg/g、纤维素酶3.3 mg/g、半纤维素酶3.3 mg/g、木质素酶1.9 mg/g;最佳酶解条件为酶解pH 5.5、酶解温度50℃、酶解时间70 min,在此条件下,多酚含量达9.21 mg/g。结构分析表明,高粱壳多酚具有多个苯环和酚羟基。抗氧化试验结果表明,高粱壳多酚具有较好的抗氧化活性。Polyphenols from sorghum husks were extracted by enzyme assisted method.Taking polyphenol content as index,the optimal enzyme dosage was determined through single factor experiments and orthogonal experiment,and then the enzymatic hydrolysis conditions were further optimized.The structural characterization and antioxidant activity of sorghum husk polyphenols were analyzed.The results showed that the optimal enzyme dosage(based on the mass of sorghum husk powder)was papain 2.1 mg/g,cellulase 3.3 mg/g,hemicellulase 3.3 mg/g,and ligninase 1.9 mg/g.The optimal enzymatic hydrolysis conditions were pH 5.5,enzymatic hydrolysis temperature 50℃,and enzymatic hydrolysis time 70 min.Under these conditions,the polyphenol content was 9.21 mg/g.Structural analysis showed that sorghum husk polyphenols had multiple benzene rings and phenolic hydroxyl groups.The antioxidant test results showed that sorghum husk polyphenols had good antioxidant activity.
