摘 要:以核桃粕为原料,采用碱提酸沉法制备核桃蛋白,利用碱性蛋白酶制备酶解产物。分别配制3种不同质量浓度(0.4、2.0、10.0 mg/mL)的核桃蛋白溶液和酶解产物溶液,与MC3T3-E1成骨细胞共同培养24、48、72 h后,对其进行形态学观察,并通过四唑盐比色法测定成骨细胞的增殖率,比较核桃蛋白和酶解产物的促成骨细胞增殖作用。结果表明:核桃蛋白和酶解产物均能显著促进MC3T3-E1成骨细胞的增殖。在不同时间点(24、48 h)的核桃蛋白和酶解产物随质量浓度的增加,细胞增殖率均逐渐升高。当核桃酶解产物质量浓度为10 mg/mL时,与核桃蛋白相比,核桃酶解产物在3个不同时间点均对MC3T3-E1成骨细胞增殖的促进效果更好,其中以培养24 h的核桃酶解产物的效果最好。Walnut protein was prepared using walnut meal as material by alkaline extraction and acid precipitation,and enzymatic hydrolysis products were prepared by alkaline proteases.Three different mass concentrations(0.4,2.0,10.0 mg/mL)of walnut protein solution and enzymatic hydrolysis product solution were prepared and co-cultivated with MC3T3-E1 osteoblasts for 24,48,and 72 h,respectively,and morphology was observed.The proliferation rate of osteoblasts was also determined by tetrazolium salt colorimetric method,and the effects of walnut protein and enzymatic hydrolysis products on promoting osteoblast proliferation were compared.The results showed that both walnut proteins and enzymatic hydrolysis products significantly promoted the proliferation of MC3T3-E1 osteoblasts.The cell proliferation rates of both walnut proteins and enzymatic hydrolysis products at different time points(24,48 h)gradually increased with the increase of mass concentration.When the mass concentration of walnut enzymatic hydrolysis products was 10 mg/mL,compared with walnut protein,the enzymatic hydrolysis products of walnut had better effect on the proliferation of MC3T3-E1 osteoblasts at three different time points,and enzymatic hydrolysis products of walnut cultured for 24 h had the best effect.
