摘 要:以ω-溴苯乙酮作为脂肪酸衍生剂,利用超高效液相色谱(UPLC)法测定双低菜籽油和结构脂质中脂肪酸组成与位置分布,探讨了流动相、柱温对脂肪酸衍生物分离检测效果的影响。经优化确定以(甲醇-乙腈)(90∶10)-水为流动相梯度洗脱,在检测波长246 nm,柱温35℃,流速0.35mL/min的色谱条件下,18 min内能完成对13种脂肪酸衍生物的分离检测。UPLC法具有操作简单,分析检测时间短,溶剂消耗少,分离度好的优点。A method was developed for the determination of fatty acids positional distribution in triacylg-lycerols of double - low rapeseed oil and structured lipid using w-bromoacetophenone as derivative reagent by ultra -high performance liquid chromatography( UPLC ). Different chromatographic parameters ineluding elution profiles, column temperature were investigated in order to optimize separation conditions of different fatty acid phenaeyl ester derivatives. A gradient elution using methanol/acetonitrile (90 : 10, V/V) -water as mobile phase at 0.35 mL/min( flow velocity), detection wavelength at 246 nm, and column temperature of 35℃ were chosen. Under these conditions, 13 derivatives of fatty acids were separated in 18 min. This method was simple, fast,little solvent-consuming and good separation degree.
