摘 要:目的:研究黑曲霉B0201液体发酵产单宁酶的条件。方法:用紫外分光光度法测定酶活,采用单因素法对黑曲霉B0201产单宁酶的工艺条件进行优化。结果:液体发酵主要产胞内酶,4%单宁酸、2.5%葡萄糖、1.25%(NH4)2SO4是产酶的最佳培养基;温度30℃,初始pH值为5.0,接种量孢子悬液1×108个/mL,装液量50mL,摇床转速140r/min是产酶的最佳工艺条件,发酵72h,单宁酶的活力最高,可达145.2U/mL。Objective: The conditions of tannase production from Aspergillus niger B0201 under submerged fermentation was studied. Methods: The activity of tannase was determined by ultraviolet spectrophotometric method. The production conditions of tannase from A. niger B0201 were optimized by single factor method. Results: Intracellular enzymes were mainly produced during submerged fermentation. The optimal medium for enzyme production contained 4% tannins, 2.5% glucose and 1.25% (NH4)2SO4. The optimal conditions for tannase production were as follows: temperature 30℃, initial pH 5.0, inoculum 1 ×10^8/ml spores, medium volume 50ml, rotating speed 140 r/min. Under above conditions, the activity of tarmase was highest and reached 145.2 U/ml after 72 h fermentation. Conclusion: Under optimal conditions, the activity of intracelllar tannase was relatively high. The fermentation processing was simple and easy to control. The results might provide important foundation and better guidance for industrial production oftannase.
