摘  要:以马铃薯淀粉为原料,采用全酶法并借以喷射液化工艺制备低DE值的麦芽低聚糖浆,采用高效阴离子交换色谱法测定所制备样品的糖分组成,离子树脂交换法从中分离制备磷酸寡糖,最后,采用薄层层析法和红外光谱法对其进行定性分析。结果表明:制备液化DE值16%,糖化DE值35%的糖液,其中,主要成分为麦芽二糖至麦芽七糖,以二糖至五糖居多。201×4强碱性苯乙烯系阴离子交换树脂分离效果好,分离条件为:洗脱液为0.4 mol/L NaCl,洗脱速度为1.4 mL/min。经红外光谱定性分析,结果显示样品为结合有磷酸基团的糖类物质。Malt-oligosaccharides with low dextrose equivalent(DE) was prepared from potato starch by using the fully enzymatic method and steam ejection liquefaction.Sample composition was detected using high performance anion-exchange chromatography,and phosphoryl oligosaccharides(POs) were separated from malt-oligosaccharides with the ion-exchange method.Finally,the POs were analyzed by thin-layer chromatography and infrared spectrum.Results: The DE values of the prepared liquefied and saccharified malt-oligosaccharides are 16% and 35%,respectively,and a majority of the product is malt-oligosaccharides of disaccharide to pentose.The optimal conditions to separate POs are using 201×4 ion-exchange resin for separation,and eluting the column with 0.4 mol/L NaCl at a speed of 1.4 mL/min.IR analysis indicates the product is phosphate groups bound saccharides.
